中枢乙酰胆硷毒蕈碱样受体结合力的测定
THE DETERMINATION OF ~3H-QNB BINDING WITH MUSCARINIC CHOLINERGIC RECEPTORS IN RAT BRAIN
【摘要】 <正> 乙酰胆硷是一种研究历史最长,在体内分布最广的神经介质,对许多生理活动都有重要的调节作用。乙酰胆硷是通过受体起作用的,因此,乙酰胆硷受体结合力的测定,对神经生理、病理和药理的研究工作都具有重要的意义。我们依据Yamamura和snyder的报导,应用~3H-QNB(~3H-Quinuclidinyl benzilate)建立了脑内乙酰胆硷毒蕈碱样(M)受体亲和力的测定方法,并结合本实验室的条件,作了一些改良,现将方法部分报告如下:
【Abstract】 This article introduces the method of affinity binding of ~3H-QNB to rat brain muscarinic cholinergic receptor in vitro.The maximal binding of ~3H-QNB (Bmax) was found to be 281 pMol/mg protein, with dissociation constant(KD) as 6.6 nM, The ratio of specific to nonspecific binding was 5:1.The binding of ~3H-QNB could be competitively inhibited by adding unlabelled agonist (acetylcholine) or antagonist (atropine) of muscarinic cholinergic receptors into the incubation medium, the muscarinic antagonist being about 100 times more effective in competition than the agonist.
