从末梢血分离淋巴类细胞一些变异因素的试验观察

【摘要】 1971年我们制成右旋糖酐-泛影葡胺液分离血中淋巴细胞取得满意效果。随后用Ficcoll-Hypaque和聚甘露醇-泛影葡胺也取得同样效果。脱纤维使血的粒细胞显著降低,本文报告无空调试验室在深秋低温10~15℃和盛夏高温30~35℃时分离淋巴细胞的技术。 我们设计了控制离心筒内微环境的温度,深秋时将离心筒预热至25℃,盛夏时10℃冰箱预冷2小时,再加脱纤维血于分离液面,离心后可得满意的分离效果。

【Abstract】 In 1971 we prepared the urografin-dextran mixture for isolation of the lymphoid cells from blood with satisfactory results. Thereafter, Ficcoll-Hypaque and manan-urografin were also investigated, the results were essentially the same as the urografin-dextran mixture. The process of blood defibrination decresed the granulocyte markedly.In this paper, we present the methad to isolate the lymphoid cells in a laboratory without air conditioner, in late autumn when the room temperature may drop to 10—15℃ and in summer when it may reach. 30—35℃. Our design is to control the microenvironment of centrifuge tubes. In late autumn, the centrifuge tubes with isolation fluid are preincubated at 25℃, and in summer they are preincubated in an ice box at 10℃ for at cleast 2 hours. Thereafter, the defihrinated blood was on the isolation fluid. After centrifugation, a satisfactory recovery rate is attained for the lymphoid cell.
