快速甲胎蛋白放射免疫测定——硫酸铵沉淀法

【摘要】 报告一个快速甲胎蛋白放射免疫测定硫酸铵沉淀方法。沉淀抗原—抗体复合物的饱和硫酸铵最终饱和度为37.5%。125I-AFP每管大约20000 CPM,马抗人AFP抗血清稀释度为1:5000,反应混合物在37℃保温4小时,3000 rpm离心15分钟。结合百分率的计算和样品中AFP的量是由标准曲线所得。文中详细叙述了操作方法,此法可测到12.5 ng/ml。

【Abstract】 Hereby a rapid method of determining serum alphafetoprotein (AFP) concentration with radioimmunoassay method is reported. The antigen-antibody complex here was precipitated with saturated ammonium sulfate solution in a final saturation 37.5%. 125Ⅰ-labelled AFP was approximately of 20,000 cpm in each tube and concentration of horse antihuman AFP antiserum was of 1:5,000. The mixture was incubated at 37℃ for 4 h. The tubes were centrifuged at 3,000 rpm for 15 min. The percent of radioactivity bound to the antiserum was calculated and the amount of AFP for different serum samples was determined from the standard curve and expressed as ng/ml. 12.5 ng of AFP per milliliter can be detected with this method. The dose-response curve covers an analytical range of 12.5 to 800 ng/ml. The method has been described in detail.
