酶联免疫吸附法(ELISA)测定乙型脑炎病人血清中特异性IgG

【摘要】 本文研究了ELISA在测定乙型脑炎病人血清中抗乙脑IgG的几个条件。比较了五种抗原。发现以高顺生株乳鼠脑悬液高速或超速离心的抗原最好。抗原的活性与附着塑料板的能力似与乙型脑炎表面血凝素有关。用这两种抗原做ELISA及血凝抑制试验,测定了31份乙型脑炎病人双份血清中乙脑特异性IgG。以抗体四倍增长作为判断标准,两种方法的结果绝大多数相符。92份单份血清,两次测定结果,符合率达93%。ELISA与血凝抑制试验所测血清乙脑特异性IgG滴度相仿。但ELISA血清用量小,方法简便。在本试验中发现Dynatech Lab.聚乙烯板略优于上海塑料三厂聚苯乙烯板。

【Abstract】 Several factors were found to affect the outcome of ELISA for the detection of specific serum lgG of Japanese B encephalitis patients studied in this paper. It was found that there were considerable differences between virus antigens produced by different strains with various laboratory procedures. Chiefly, the adhering ability on the plastic surface of antigens prepared from infected suckling mouse brain tissue with virulent Japanese B encephalitis virus strain ’Kao’ is greater than that produced either by the attenuated strain No 2—8 or the virulent strain ’Kao’ cultured on hamster kidney cells. It was shown that infectivity of these two kinds of virus is almost equal. The TCD50 of suckling mouse brain virus and hamster kidney tissue culture grown virus are 8.3 and 8.0 respectively. However, haemagglutinating titre of tissue culture antigen is much lower (1:8) than the brain antigen (1:40960). So, it was assumed that the adhering ability of viral antigen in ELISA is correlated with haemagglutinin titre.2 single sera specimens and 31 paired sera from Japanese B encephalitis patients were tested both by ELISA with suckling mouse brain antigen and haemagglutination inhibition test. The results indicated that these two methods correlated rather well. Especially the 92 single sera were tested twice by both method, Indeet they have definite agreement. The sensitivity of these two methods is similar to each other However ELISA is more covenient and requires only smaller amounts of serum. The results with two kinds of plastic plates were also compared. It was shown that Dynatech laboratory polyvinyl plates is somewhat superior to Shanghai polystyrene plates. However good results could also be obtained by the latter when optimal concentration of antigen was selected
