小麦种子萌发时乙醇脱氢酶的变化及其同工酶的诱导形成
ACTIVITY OF ALCOHOL DEHYDROGENASE AND THE INDUCED FORMATION OF ALCOHOL DEHYDROGENASE ISOZYMES DURING GERMINATION OF WHEAT SEEDS
【摘要】 小麦种子在空气条件中萌发时,乙醇脱氢酶随发芽时间延长而降低。在低氧和高二氧化碳浓度中萌发的种胚,酶活性增加的幅度随厌氧的程度和CO2浓度的增加而上升。用1%乙醇处理小麦种子,并令其在乙醇中发芽三天,对照的ADH活性几乎完全丧失,处理仍有很高活性,约为对照的10倍。在上述不同条件下萌发的种胚,ADH同工酶也显示不同的区带,其中用高浓度CO2处理,在偏阳极一端比对照多两条带,CO2浓度愈高这两条带愈明显。这些结果表明,种子萌发时ADH活性及其同工酶的变化与组织中内源乙醇的浓度有关。说明乙醇是ADH的天然诱导物。ADH为一种诱导酶。
【Abstract】 In the present work the wheat seeds were used as the main materials for the investigation of the activity of alcohol dehydrogenasc (ADH) and its isozymes. It was shown that the ADH activity decreased during seed germination under aerobic condition, but increased under low oxygen and high carbon dioxide partial pressures. A relatively higher alcohol dehydrogenase activity was also found in the embryo from seeds germinated in solution of one per cent ethanol.The ADH isozymes appeared to be different under various environmental conditions. After applying high carbon dioxide to the seeds, two new bands of ADH isozymes appeared on gel electrophoresis.These results suggest that the alcohol dehydrogenase activity and the change of ADH isozymes in the embryo during germination is closely related to the endogenous concentration of ethanol within the plant tissue.The above results indicated that alcohol is the natural inducer of ADH in germinating seeds. As the formation of ADH is induced by alcohol, we suggest that ADH is an induced enzyme.
