植物叶绿素-蛋白质复合物的研究——Ⅰ.阳生植物向日葵、阴生植物—叶兰的叶绿素-蛋白质复合物

【摘要】 阳生植物向日葵的叶绿体囊状体膜,用SDS-聚丙烯酰胺凝胶电泳最多可分离出8条含叶绿素带:复合物Ⅰa、Ⅰb、Ⅰc、Ⅱa、Ⅱb、Ⅳ、Ⅱc和游离色素去垢剂复合物Ⅲ。从阴生植物一叶兰只分离出6条含叶绿素带,属光系统Ⅰ的只有一条带(Ⅰc)。向日葵的复合物Ⅳ,从电泳迁移率、吸收光谱、光密度扫描等看来,如最近有的实验室所提出的那样,是与PSⅡ反应中心有关的。一叶兰复合物X的电泳迁移率与向日葵复合物Ⅳ相似,但它不但含Chl a还含有Chl b。它在室温下680nm的荧光发射与复合物Ⅳ十分相似,在所有这些复合物中,它们二者的发射最强。阳生植物(向日葵、菠菜、莴苣)叶绿体的Chl a/b值比阴生植物(一叶兰、广东万年青、吊兰)高,阴生植物(一叶兰)LHCP复合物的量大于阳生植物(向日葵)。并初步看出,阴生植物(一叶兰)LHCP复合物的Chl a/b值也低于阳生植物(向日葵)的相应复合物。

【Abstract】 The chlorophyll-protein complexes of thylakoid membranes of the sun plant sunflower chloroplasts can be resolved by SDS-PAGE into up to 8 chlorophyllcontaining bands: Complexes Ⅰ_a, Ⅰ_b, Ⅰ_c, Ⅱ_a, Ⅱ_b, Ⅳ, Ⅱ_c and Ⅲ (free pigments complexed with SDS).From the shade plant, Malaxis monophyllos (L.) Sw. only 6 chlorophyll-containing bands, including one CP I band (I_c), can be resolved.Electrophoretic mobility,absorption spectra and densitometric scannings suggest that complex IV of sunflower is associated with the reaction center of PS Ⅱ as several laboratories claimed recently. Complex X of M. monophyllos has similar mobility as complex Ⅳ, but contains Chl b in addition to Chl a. Of all these chlorophyll-protein complexes,complex X emits the strongest 680 nm fluorescence at room temperature just as complex Ⅳ.In addition to the fact that Chl a/b ratio of chloroplasts from sun plants (sunflower, spinach and lettuce) are higher than that of shade plants (M. monophyllos, Aglaonema modestum Schott and Chlorophytum comosum (Thunb.) Jacques), we observe that the quantities of LHCPs of the shade plant (M.monophyllos) are larger than those of the sun plant (sunflower). It seems that Chl a/b ratio of LHCP complexes of the shade p/ant (M. monophyllos) may be lower than that of the sun plant (sunflower).
