【Abstract】 This paper reports on the pharmacokineti cs of naproxen in laboratory rabbits and human subjects.The naproxen content in serum was determined spectrophometrically as 233 nm, after extraction by organic solvent and counter-extraction by Xa-OH.A dose of 10mg/kg body weigt of naproxen sodium was administered intravenously to the rabbits. Serum concentration-time curve was shov,-n to fit a two compartment open model with mean half-life (T(1/2)β) of 8.05 hr and mean apparent distribution volume in central compartment ( Vc ) of 0.1503L.Two 250mg capsules of naproxen was given orally to 5 healthy volunteers. Serun concentration-time curve was assumed to fit a one compartment open model with first order absorption. The pharmacokinetic parameters calculated by the data from 4 persons were: mean absorption rate constant ( ka ) 1.6613 hr-1, mean peak time ( Tmax ) 2.432 hr, mean elimination rate constant ( K ) 0.03243 hr-1, half-life ( T(1/2) ) 21.81±3.86 ( SD ) hr. The ka and Tmax values of another case differed significantly from that of the above 4 cases.更多还原
