甜椒花药培养的初步研究
A PRELIMINARY REPORT ON THE STUDY OF POLLEN PLANTS OF SWEET PEPPERS (CAPSICUM FRUTESCENS L. VAR. GROSSUM BAILEY)
【摘要】 花药培养实验共用十九个甜椒品种,在十六个品种中曾获得胚状体,但由于多数胚状体不能正常发育,仅有“四方头”和“旅大”两个品种的花药在附加0.25毫克/升萘乙酸(NAA),1毫克/升激动素,50毫克/升单核苷酸的NT_H培养基(蔗糖3%)上产生了比较正常的花粉胚状体。将其转移到含有0.1毫克/升吲哚乙酸(或0.001毫克/升NAA),2─4毫克/升激动素,4毫克/升6—苄基嘌呤(或10%椰子乳)的NT_H培养基上,花粉胚状体进一步发育为单培体植株(n=12)。花粉胚状体的诱导率因品种而异。低浓度的萘乙酸有利于胚状体的形成,但是不同品种在形成花粉胚时所需要的萘乙酸浓度不同,将获得的花粉植株用0.2%的秋水仙碱液进行处理24小时,其中染色体获得加倍的植株,能开花结果。
【Abstract】 Of 19 cultivars of sweet peppers used for anther culture 16 of them gave rise to embryoids, but normal pollen embryoids were obtained only from two cultivars, i.e. "Square Head" and "Lu-tai" ,whereas the embryoids from o-ther cultivars could not develop normally.The basic medium(NTH) was supple-mented with 0.25mg/l NAA,lmg/l kinetin, 50 mg/1 nucleotide and 3% sucro-se. The pollen embryoids were further developed into haploid plantlets(n= 12) after transfer to NTH medium supplemented with 0.lmg/1 IAA (or 0.00lmg/1 NAA) , 2-4mg/l kinetin, 4mg/l 6-BAP (or 10% fresh coconut milk).The frequency of embryoid induction varies with the cultivar. LOW concentrations of NAA are beneficial to embryoid formation but the optimum concentra-tion varies with the cultivar.The haploid plantlets were then treated with 0.2% cochicine solution or 24 hours. Among them, only the diploidized plants gave flowers and set seeds.
