【Abstract】 An exogenous RNA dependentcell-free protein synthesizing systemfrom wheat germ extract (S26) hasbeen discribed.This system is pre-pared by simply passing through DEAE-cellulose (Cl-form) in the presence of0.3 M KCl and then by dialysingagainst a buffer containing 20%glycerol.The advantages of this cell-free system are as follows:1.The conventional method wassimplified by passing through DEAE-cellulose instead of ultracentrifuga-tion.2.The endogenous tRNA,mRNAand amino acids have been removedalmost completely,so that it becomesan exbgenous RNA dependent cell-free protein synthesizing system.3.The protein synthesizing acti-vity of the final preparation can bepreserved much longer in deep freezer(-20℃) than the system preparedbefore.
