不同品系金鱼间细胞核的多代移植

【Abstract】 Different nucleo-cytoplasmic com-binations were made between threevarieties of goldfish by means of tran-splantation of blastula nucleus of onevariety to enucleated mature oocyte ofanother.The varieties used were theRed Dragon with double caudal finand protruded eyes;the Red Casas-sius with single caudal fin and normaleyes;and the Eggfish with double caud-al fin and normal eyes.The indifidualsfrom these nucleo-cytoplasmic combina-tions not only developed normally butalso reached sexual maturity.61 adult fishes were obtained fromsuch combinations (N1).Their pheno-typic expressions were fundamentallyof the nuclear donor type,E G Whenthe nucleus of the Red Dragon com-bined with the cytoplasm of the RedCarassius,the resulting fish had doublecaudal fin and protruded eyes.Thesame was true for the reciprocal com-binations.When the N1 male was matedwith a female pf ots nuclear donorvariety:Red Dragon nucleus to RedCarassius cytoplasm ♂×Red Dragon ♀,gave off-spring of the nuclear donorphenotype.e.g.the double tailed N1male mated with a Red Dragon female,gave off-spring all with double tail.This indicated that the Red Dragonnucleus,although duplicated hundredof times in the single tailed cytoplasmwas not effected by the host in regardingto its phenotypic expression.The blastula nucleus of these N1×Red Dragon was again transplantedinto enucleated mature oocyte of theRed Carassius,the resulting fish (N2)developed either single or double tailfin,the former was quite numerous.This indicated that the nuclear factorof the N1 although not altered by theforeign cytoplasm as expressed by ma-ting with female of its own nucleardonor variety had become more plastic and was more easily influenced whenit was again introduced into the cyto-plasm of the sihgle tailed variety.The effect of cytoplasm upon thephenotypic expression was extremelyevident in such twin or triplet cases,raised from nucleo-cytoplasmic com-binations with nuclei taken from thesame donor and different cytoplasmiccontent.They had tail of variousforms:one single and the other doubleor two single and the third double orvice versa.When the single tailed N2 malewas again mated with a double tailedRed Dragon female,0.4% of the off-springs showed typical single tail and23% of them had atypical single tail:single tail fin plus a small piece.Whensuch blastula nuclei were transplantedinto enucleated Red Carassius eggs(N3) the percentage of the single tailedones increased still higher.The singletailed ones of N4 were even more nu-merous than that of N3.The effect of cytoplasm upon thephenotypic expression was quite clearin this experiment.The nucleus sho-wed actual alteration as revealed byits phenotypic expression after trans-plantation for four generations.
