淡色库蚊对敌百虫抗性的研究——水解酶同敌百虫抗性关系
STUDIES ON THE RESISTANCE OF DIPTEREX-RESISTANT MOSQUITO CULEX PIPIENS PALLENS Coq.——ON THE RELATIONSHIP BETWEEN HYDROLASE AND RESISTANCE
【摘要】 本文利用离体水解酶测定和聚丙烯酰胺圆盘电泳技术研究了淡色库蚊(Culex pipiens pallensCop.)对敌百虫抗性和水解酶的关系。实验结果表明:(1)抗性品系的羧酸酯酶活力远比敏感的大,并随着对敌百虫抗性水平的升降而增减。(2)酯酶同功酶谱表明,抗性品系有一染色强度很深的特征性羧酸酯酶带E5,该带在抗性和敏感品系的杂交子代F1中也发现,但强度不如抗性品系,这同F1代的抗性水平下降相平行。(3)无论胆碱酯酶或是磷酸酯酶活力,在二品系中未见到明显差异。抗性和敏感品系的胆碱酯酶对敌百虫和敌敌畏的敏感度相似。
【Abstract】 Polyacrylamide gel electrophoresis and enzymatic determination in vitro were used to demonstrate the relationship between hydrolase activity and resistance. The results are briefly stated as follows:(1) The earboxylesterase activity were considerably stronger in the resistant strain than in the susceptible strain. The decrease of Dipterex-resistance in the resistant strain reared in an insecticide-free environment was accompanied by a corresponding drop in earboxylesterase activity, and the earboxylesterase activity in the resistant strain increased with the development of Dipterex-resistance.(2) Esterase zymograms from polyacrylamide gel electrophoresis revealed both strains had similar patterns except the E8 band which was much more intense in the resistant than in the susceptible strain, as judged by its hydrolysis of α-and β-naphthyl acetate. The results of eleetrophoresis scanning showed earboxylesterase increase mainly resided in this band. This band can also be found in the F1 strain (R × S) but the activity is less than in the resistant strain, which was in line with the decrease of resistance in the F1 strain.(3) No difference was observed in phosphatase and cholinesterase, not only in enzymatic determination in vitro but also in the esterase zymograms. The inhibition of cholinesterase of both strains by Dipterex and DDVP was similar.
