固相放射免疫法检测血清中HBcAg
DETERMINATION OF SERUM HBcAg BY SOLID PHASE RADIOIMMUNOAY(SPRIA)
【摘要】 <正> 乙型肝炎的完整病毒(HBV)是Dane颗粒,早在1970年已被Dane证实。被HBV感染后的患者血清内存在这种Dane颗粒,用去垢剂Tween 80处理,可使Dane颗粒裂解为内核和外壳两部分。其外壳即表面抗原(HBsAg),是由蛋白质、脂质和碳水化合物组成,人体被感染后肝细胞浆内可大量复制HBsAg,机体即可产生相应的抗体,即抗-HBs,国内外对此系统研究较多并先后建立了敏感的检测方法。其内核部份是HBV的第二抗原抗体系统,称核心抗原抗体系统(HBcAg、抗-HBc)。不少报导认为抗-HBc的出现是代表病毒的复制,而肝细胞核内的HBcAg则更是
【Abstract】 Dane’s particles were precipitited in the serum by specific antibody followed by thetreatment with NP-40, then exposed to hepatitis B core antigen (HBcAg) and tested for theHBcAg by the SPRIA. A procedure of determining HBcAg in the serum and various in-fluencing factors and serum samples of 20 healthy donors and 20 carriers of HBsAg werepresented. It is found that the value (test sample/control sample cpm) for the 20 healthy donorswas 0.39--1.47 (<2. 1), the value for the 10 asymptomatic carriers with higher HBsAgtiter (7 with HBeAg(+). 3 with HBeAg(-) anti--HBe(-)) was 5.57--34.98(>2. 1) how-evev 10 HBsAg (+) with anti--HBe (+) was 0.31--1.73. These results are parallel withDNAp activity, therefore SPRIA has higher specificity and sensitiveness as well as the bet-ter repetiveness. It is sensitive index for the HBV infection and prognesis.
