骨巨细胞瘤体外培养的显微缩时电影观察——Ⅰ.多核巨细胞和间质细胞的体外生长行为
TIME LAPSE CINEMICROGRAPHIC OBSERVATIONS ON GIANT CELL TUMOR OF BONE CULTURED IN VITRO
【摘要】 <正> 我们应用相差显微缩时定格摄影技术,先后观察了9个骨巨细胞瘤标本在体外培养中多核巨细胞和间质细胞的生长行为,包括:细胞从组织块移出的过程,细胞的一般动态,细胞死亡崩解,间质细胞的分裂及多核巨细胞的自然融合。本文的图片系从影片中选出作为示例。
【Abstract】 Time lapse cinemicrographic observations on 9 giant cell tumors of bone cultured invitro were undertaken in the past two years. Tumor tissue were cut into small pieces and explanted on a cover glass fixed to a win-dow of the culture flask specially made for this purpose. Observations began 8-24 hoursafter explantation. Growth of fibroblast-like stromal cells and multinucleated giant cells(M. G. Cells) took place in all 9 cultures. In general, the former cells migrated out fromthe explant earlier than the latter. Features of migration of M. G. cells were remarkable. At first, a tongue-like cyto-plasmic process with a ruffled or semi-circular border protruded out from the edgeof the explants. It took about 10 hours for a M. G. cell to move out completely. Theseisolated free M. G. cells usually showed active ameboid movements. The veil--like cytoplasmwith an undulating membrane incessantly changed their contours. The nuclei in M. G. cellsmostly collected in the central portion and only occassionally in the peripheral zone of thecell. Vacuoles of varying sizes were frequently found in the cytoplasm, but the majority ofthem resolved in a short time. An outspread M. G. cell at times contracts, forming a sphe-rical or oval cell, and then, the nuclei were no longer distinctly seen. In addition, vigoruos pinocytosis and phagocytotic activity of cells of both kinds werevisible. Mitosis of the stromal cells have been recorded, but the same process has not yetbeen found in the M. G. cells in the present study. Gellular contact accompanied by exchange or delivery of intracytoplasmic granules thro-ugh the inter-cellular gap junction has been shown. This phenomenon probably representscellular direct communication. The most interesting finding that had been observed wasnatural fusion of two M. G. cells. This is hitherto undescribed. But death of the fused giantcell occurred in 24 hours. The stromal cells soon gathered around this dead giant cell andvigorously phagocytized the remains which was cleared up in 7 days. Cinemicrographic provids visual records, in motion, of the growth behavior of the cellu-lar elements of giant cell tumor of bone. The present communication is the initial part ofour work and further reports will be forthcoming.
