离体棉花胚珠的纤维生长和幼苗形成

【摘要】 受精棉花胚珠培养在Beasley培养基上能长出纤维,两周后纤维停止生长,偶然可达三周。赤毒酸(GA3)或GA3加吲哚乙酸(IAA)能促进受精或未受精胚珠纤维的伸长。离体培养的棉胚珠能形成胚。胚珠培养中加入GA3后,使胚的出现率下降。从培养的胚珠中剥离的胚先培养在加有2mg/1激动素(Kinetin)的Beasley固体培养基上(0.6%琼脂),有利于子叶的转绿和展开,再转移到Radin离体棉根培养基或Kasperbauer的生根培养基上,有利于根系的生长,形成幼苗。幼胚直接培养在Radin培养基上也能成苗。

【Abstract】 Cotton ovules were removed aseptically from ovaries before or after anthesis and cultivated by floating on the surface of the liquid Beasley’s medium, fibers would then be produced. The growth of the fiber usually stopped after two weeks but occasionally continued for three weeks. Plant growth hormones such as GA3 or GA3 plus IAA promoted both the fertilized and the unfertilized ovules to produce fibers.The excised ovules produced embryos on the culture medium, but after the addition of GA3 into the medium, the frequency of embryo formation decreased. By shelling off the coat of ovules, cultivating the embryos on the solid Beasley’s medium (0.6% Agar+2mg/l kinetin), cotyledons would become green in color and expanded. Transfer of the embryo to Radin’s medium or Kasperbauer’s medium was favorable for the formation of roots and seedlings, seedlings could also be induced by directly cultivating the young embryos on the Radin’s medium.
