肿瘤的化学治疗ⅩⅩⅩⅣ——DL-N-二氯乙酰基-5-硝基-色氨酸及其有关化合物的合成

【摘要】 <正> 作者之一曾合成DL-N-二氯乙酰基-7-硝基-色氨酸(Ⅰ_a)和DL-7-硝基-色氨酸盐酸盐(Ⅱ_a),及其相应乙酯即DL-N-二氯乙酰基-7-硝基-色氨酸乙酯(Ⅰ_b)和DL-7-硝基-色氨酸乙酯盐酸盐(Ⅱ_b),经组织培养筛选表明Ⅰ_a和Ⅰ_b对Hela瘤细胞有明显抑制作用。为进一步研究硝基位置对肿瘤抑制作用的影响,本工作又合成了其相应的5-硝基位置异构体,DL-N-二氯乙酰基-5-硝基-色氨酸(Ⅰ_c)和DL-5-硝基-色氨酸盐酸盐(Ⅱ_c),及其相应的乙酯Ⅰ_d

【Abstract】 In the present work, the compounds DL-N-dichloroacetyl-5-nitrotryptophan (I_c), DL-5-nitro-tryptophan hydrochloride (Ⅱ_c) and their ethyl esters (Ⅰ_d and Ⅱ_d) were prepared and found to be less effective against Hela cells than their corresponding 7-nitro analogs (Ⅰ).5-Nitrogramine (Ⅴ) was condensed with diethyl formamidomalonate and dimethyl sulphate in the presence of sodium alcoholate, affording diethyl α-formamido-α-(5-nitroindolyl methyl)-malonate (Ⅵ). The latter was hydrolyzed in concentrated hydrochloric acid to give 5-nitrotryptophan hydrochloride(Ⅱ_c), from which the desired product, DL-N-dichloroacetyl-5-nitrotryptopban (Ⅰ_c), was obtained by acylation with dichloroacetylchloride in dilute sodium hydroxide. The ethyl ester of DL-5-nitrotryptophan (Ⅱ_d) was obtained by esterification of Ⅱ_c with absolute ethyl alcohol saturated with gaseous hydrogen chloride and was then treated with dichloroacetyl-chloride in the presence of aqueous sodium acetate or in boiling benzene.
