綿羊布氏桿菌病血清學診斷的研究——Ⅰ.10%鹽水代替生理鹽水作凝集反應稀釋液的研究
STUDIES ON THE SEROLOGICAL DIAGNOSIS OF OVINE BRUCELLOSIS Ⅰ. OBSERVATIONS ON THE SUBSTITUTION OF NORMAL SALINE BY 10% SALINE AS A DILUENT IN TUBE AGGLUTINATION TEST
【Abstract】 In conducting the experiments, 16 artificially infected sheep, and 48 reactors in addition to 6958 sheep taken from infected and noninfected farms were subjected to periodic examinations for brucellosis by serological, bacteriological, and pathological methods. Preliminary results were summerized as follows. 1. When examined by ordinary tube agglutination test with normal saline (0.85% NaCl) as diluent, the agglutinating titres of the sera of both naturally and artificially infected sheep showed marked fluctuations at various periods; at times the positive reactions gave place to negative ones. But when 10% salt solution was substituted for the normal saline as a diluent, such fluctuations did not occur; serum titres were stable, generally somewhat higher than those obtained by the ordinary test, and were very much in accord with the results of the complement-fixation test. 2. When 6958 field samples of sera were examined by the agglutination tests using both 0.85% and 10% saline solutions as diluents and by the complement-fixation test, the rate of positive reactions (including the suspicious reactions) given by the ordinary agglutination test was 2.8—4.2%, that given by the test using 10% saline as diluent 5.4—8.5%, and that given by the complement-fixation test, 7.1%. The 10% saline test covered all the reactors picked out by the ordinary test, and also 2.9% of reactors missed by the comple-ment-fixation test. However, the latter test detected in addition a small portion (1.6%) of reactors missed by the 10% saline test. The results of pathological and bacteriological exminations of both naturally and experimentally infected sheep reported in this study confirmed the value of practical application of the observations made by P. A. Trilenko in 1954. The authors strongly recommended the 10% saline as a substitute for the normal saline which was ordinarily used as a diluent in the tube agglutination test for the diagnosis of ovine brucellosis,
