钝齿棒状杆菌噬菌体的鉴定及其核酸的分离和测定

【摘要】 对生产中广泛使用的钝齿棒状杆菌的噬菌体,进行了血清学分型,区分为两种血清型,并按代表株B271和B 275的编号分别定名。这两株噬菌体的寄主范围十分严格,仅限于钝齿棒状杆菌及其相近的菌株。60℃处理后基本上失活。B275噬菌体对pH值的稳定性较B271噬菌体稍强,但二者对柠檬酸钠的反应截然不同,在试验浓度范围内,前者表现稳定,而后者不稳定。一级生长曲线试验表明B 271和B 275的潜伏期分别为78和65分钟,裂解量分别为44和54。B 271和B 275的DNA G-C%分别为57.80和64.87。此外,进行了电镜下的形态观察。

【Abstract】 L-Glutamic acid-producing bacteria——Corynebacterium crenatum AS 1.542and B9 are the current productive strainsin China. Two serotypes of phage de-singnated as B271 and B275 attackingC. crenatum were characterized by host-range, one step growth experiment andG-C% of their DNA. Neither of themis capable of infecting selected bacterialstrains other than C. crenatum and itsclosely related strain T6-13. Their la-tent periods were 78 and 65 minutes andburst sizes 44 and 54, respectively. TheT_m of B271 and B275 DNAs were 93°and 95.9℃, respectively. Their G-C%calculated by Doty’s equation were 57.80and 64.87, respectively. In sodium citrate test a striking dif-ference between B271 and B275 wasfound with B271 highly sensitive to di-valent cations for adsorption, while B275was rather insensitive. This fact mustbe considered in the control of phage in-fections with chelating agents such asoxalic acid or citric acid. It is provedthat this test is also useful in the charac-terization of phages of L-glutamic acid-producing bacteria. Both phages are more stable at pH7.0-11.0, but phage B275 is also stableat pH 5.0-6.0. The survival rates ofthese phages decline rapidly at 60℃. Electron micrographs of these pha-ges have shown that they are tadpole-liketype with a flexible tail. They are simi-lar in outline but quite different insize.
