苏芸金杆菌噬菌体的形态观察及噬菌体GP-10在寄主细胞内增殖
MORPHOLOGICAL OBSERVATIONS OF BACTERIOPHAGES OF BACILLUS THURINGIENSIS AND THE DEVELOPMENT OF PHAGE GP-10 IN BACTERIAL HOST CELLS
【摘要】 本文对山东大学生物实验厂几年来从周围环境和发酵裂解液中分离的10株噬菌体进行了寄主范围(包括9个血清型共55株苏芸金杆菌)的测定和电子显微镜观察和比较。根据其形态,可归纳为三种不同的类型。同时以噬菌体GP-10,在感染寄主细胞后的不同时间进行了电子显微镜观察。结果表明噬菌体核酸在侵入细胞后,寄主细胞核区明显扩大,并在扩大的核区复制,逐渐出现电于密度较大的噬菌体颗粒,数量逐步增加,至增殖后期,几乎充满了整个扩大了的核区。最后导致寄主细胞壁破裂而释放出成熟的噬菌体。其数量估计每个细胞接近1000个。
【Abstract】 10 isolates of bacteriophages ofBacillus thuringiensis were obtained fromthe workshop environment and phagoly-sates of fermentation tanks in Biologicaland Experimental Factory of ShandongUniversity in the past few years. Analysisof host range was made by using 55strains of Bacillus thuringiensis belongingto 9 standard serotypes. Morphologicalobservations were carried out underelectronmicroscope. According to theclassification system for bacteriophagesissued by International Committee onNomenclature of Viruses (ICNVφ), thebacteriophages mentioned above are classi-fied on morphological bases into 3 groups. A strain of Bacillus thuringiensis var.galleriae was infected with specific phageGP-10. Ultrathinsections of infected bac-terial cells were made and examined underelectronmicroscope. After adsorption ofthe phage particles on bacterial cell walland penetration of phage nucleic acid, thefirst sign of phage multiplication as seenin the section is the enlargement of thenucleoid area. As the phage particles arebeing assembled, electron dense particleswith hexagonal profile appear first in thenucleoid areas. They increase then innumber and the distended nucleoid areasare eventually full of regularly arrangedphage particles. At the final stage ofmultiplication, the mature phage particlesare released into the surroundings due tothe burst of the cell wall, and the ghost ofthe bacterial cell wall is left behind. Itis estimated that there are about 1,000phage particles per cell.
