利用光学显微镜观察泡桐丛枝病株中的类菌质体

【摘要】 应用Feulgen染色切片法检测了泡桐丛枝病株中类菌质体的存在。所得结果表明Kartha等1975年报道的方法是有效的。在病株不同部位取样,韧皮部或叶脉切成0.5厘米大小固定在Helly氏固定液中。染色操作与Kartha等相同。切片在光学显微镜下检视,类菌质体呈现暗色斑块或斑点,凝聚在树木细胞中心或细胞边缘,植物细胞作为背衬呈绿色。

【Abstract】 Feulgen staining method was appliedto detect the existence of MLO in Paw-lownia with witchhs’ broom symptoms.As our results show, this method is aseffective as the method reported by Kar-tha et al. in 1975. The phloem of the trunk or rootsfrom diseased tree and the main veins ofdiseased leaves were used as materials.They were cut into 0.5cm in size andfixed in Helly’s fixative. All the sec-tioning and staining methods were afterKartha’s. After staining sections wereexamined under ordinary light micro-scope. MLO appeared as dark dots orapots concentrated either in the center ornear to the cell wall, while the plantcells stained green.
