应用不完全抗体检查法診断布魯氏菌病的实驗研究 Ⅰ.豚鼠和羊抗球蛋白血清制备和不完全抗体检查方法的研究
EXPERIMENTAL STUDY ON THE USE OF INCOMPLETE ANTIBODIES FOR THE DIAGNOSIS OF BRUCELLOSIS——Ⅰ.PREPARATION OF ANTIGLOBULIN SERUM AND STUDY OF THE METHOD
【摘要】 <正> 文献报导痢疾、伤寒、结核等传染病患者血清中可以发现不完全抗体。这一类型抗体在性状上与完全抗体(如一般凝集素等)不同,主要表现在出现的时期、耐热性、在生理盐水中的反应、白蛋白溶液中的反应、蛋白化学性状、电泳分析、分子量等方面。一般公认在检查不完全抗体方法中以Coombs试验最精确和敏感。最近十年来欧美、日本等国试验以这种方法来诊断人、畜布鲁氏菌病;尤其是疾病的早期和慢性期的诊断。例如。Renoux(1957)用羊型布鲁氏菌感染的山羊实验研究表明:利用Coombs实验可在感染后的12周内,在全部山羊血清中发现不完全抗
【Abstract】 It is well known that incomplete antibodies may be present in patients sufferingfrom various infectious diseases,and Coombs test has been generally employed.In recentdecade,incomplete antibodies have been examined in the sera of brucellosis,and Changhas reported its use among his cases.In the present report,the method recommendedby Milgrom and his coworkers has been adopted for the preparation of the antiglobulinserum,in which the red blood cells from the rabbit,sensitized with serum to be im-munized,were used as antigen for the immunization of the rabbits.Before use,the serawere adsorbed with the rabbit red blood cells to eliminate the non-specific agglutinins.The sera obtained showed a precipitation titre of 1:16000 against the sheep serum and1:8000 against that of the guinea pig. The Coombs test was carried out in 3 steps.In the first place,the usual Wright’stest was carried out with the exception that the volume of antigen and serum was dou-bled.In the second step,all those tubes in which no agglutination had occurred werecentrifugalized and the sediments washed three times with normal saline.Then one dropof the washed sediment was placed on the slide,to which one drop of undiluted anti-globulin serum was added.The two drops were well mixed,and the appearance ofminute particles indicates positive reaction.Finally,the tube method of Coombs test wasalso carried out.To the washed sediments prepared above,0.5 ml normal saline and0.5ml of 25—50 fold diluted antiglobulin serum were added.The mixtures were in-cubated at 37℃ for 24 hours.As controls,sediments from mixtures of antigen andnormal serum,and of antigen alone plus antiglobulin serum were employed.The resultsshowed that the test to be specific and sensitive in a limited number of specimens underthe laboratory conditions.
