白喉杆菌在深层培养中氨基酸利用与产毒的关系

【摘要】 <正> 关于白喉杆菌的深层培养,Linggood,Edwards等人曾有过一系列的报导。Linggood等(1955)用80升铝罐,胰酶消化牛肉培养基,产毒为200—259Lf/毫升;Edwards(1958)用番瓜酶消化牛肉培养基产毒最高为305Lf/毫升;该氏等于1960年又提高到345Lf/毫升,井进行了酮酸代谢、生长及铁的利用与产毒关系的探封。 1962年,我们用Pope氏培养基,Linggood及Fenton(1947)胰酶消化液配方,

【Abstract】 Using PW 8 Weissensee strain and Pope’s tryptie digest medium in submerged cul-ture,a toxin yield of 150—250 Lf/ml has been obtained.At that time nearly all theutilizable amino acids have been exhausted,and the toxin yield could be augmentedwhen ammonium glutamate was supplemented from time to time during later periods ofcultivation.However,the results were inconsistent unless cystine was supplemented atthe same time.Further,a still higher yield (i.e.,410 Lf/ml) could be reached,whenMueller’s solution B was supplied during this period. The log phase growth of Weissensee strain in Pope’s medium in submerged culturelasted for about 18—24 hours.When cystine and ammonium glutamate were added atabout the 24th hour,the log phase was prolonged,or a second log phase appeared.It issuggested from the experimental results that the increase in toxin yield was due to thegrowth promoting effect of the amino acids added.
