稻属植物的组织培养和细胞分化及其遗传变异的研究——Ⅰ.粳型杂交水稻茎尖愈伤组织的悬液培养与细胞分化及其再生植株的各种变异研究
STUDIES ON TISSUE CULTURE AND CELL DIFFERENTIATION,INHERITANCE AND VARIATION IN ORYZA——Ⅰ.SUSPENSION CULTURES OF SHOOT APEX CELLS AND VARIATIONS OF CELL DIFFERENTIATION AND REGENERATED PLANTLETS IN THE HYBRID RICE OF ORYZA SATIVA
【摘要】 本文报道利用粳型杂交水稻苗端诱导形成的愈伤组织进行悬液培养试验,获得了初步成功。试验系采用MS 和N6基本培养液,另加2,4-D 1—2毫克/升,IAA0.5毫克/升,水解乳蛋白500毫克/升,蔗糖3%。结果均能生长。当愈伤组织生长至直径2—3毫米大小时,将其转移到固体再分化培养基上,使其再生形成绿苗。再分化培养基为MS 或N6基本培养基另加激动素2毫克/升,6-BA 1毫克/升,水解乳蛋白800毫克/升,蔗糖3%以及琼脂1%。经过了多次悬液继代培养以后,发现在各个“愈伤无性系”之间,于细胞形态和器官分化方面,均发生了各种各样的变异。本试验的目的,在于探索能否为利用悬液培养方法保持杂交水稻的杂种优势以及进行工厂化生产秧苗开辟道路。
【Abstract】 This paper givesa preliminary result on the success of some suspension culture experiments with the calli induced from the shoot apices of thehybrid rice in Oryza sativas sbp.Keng.The basic culture media used wereMS and N6 with the addition of 2,4-D 1-2mg/l,IAA 0.5 mg/l,lactalbu-min hydrolysate 500 mg/l,and sucrose3%,and the cultures all grew well.When the calli were transfered fromthe suspension cultures to the rediferen-tiation medium,MS or N6 basic mediumwith the addition of Kinetin 2 mg/l,6-BA Img/l,lactalbumin hydrolysate800mg/l,sucross 3%,and agar 1%,thegreen plantlets then grew up.After6 to 7 times of suspension subcultures,it was observed that variations occur-red in cell morphology and orgen dif-ferentiation between the various calli-clones.The purpose of this experim-ent is to explore the possibillty whetherthe suspension culture method maymaintain the hybrid vigor of the hynridrice,and to explore the way by whichthe production of the rice seedling(?)may be industrialized.
