免疫核糖核酸传递对人脑胶质瘤碱性蛋白抗原的细胞免疫反应研究

【摘要】 对用人脑胶质瘤碱性蛋白(GBP)免疫的38只豚鼠和23只对照组豚鼠进行了巨噬细胞电泳试验(MEM),其巨噬细胞电泳减缓率免疫组为6.13±1.06%,对照组为0.96±0.90%;经统计学处理,两组间有显著区别,表明GBP能激发豚鼠的细胞免疫反应性。从三批免疫组和三批对照组豚鼠的脾脏和淋巴结组织中分别提取I-RNA和N-RNA。将49人次健康人的淋巴细胞分别与上述RNA温育,则经I-RNA处理的淋巴细胞能对GBP产生7.73±0.97%的巨噬细胞电泳减缓作用,而经N-RNA处理的仅产生1.29±0.81%的减缓作用,经统计学处理,两组间巨噬细胞电泳减缓率有非常显著的区别,说明GBP免疫组的I-RNA能超越种间界限,使人淋巴细胞获得具有对GBP起免疫反应的能力。

【Abstract】 Previous studies have shown thatlymphocytes from patients with malig-nant disease, when encountering abasic protein fraction isolated fromhuman glioma (GBP), display a slowingeffect to macrophage electrophoreticmobility (MEM). In this report MEM tests havebeen performed with lymphocytes ob-tained from 38 guinea-pigs immunizedwith GBP and from 23 controls.Macrophage percentage slowing inMEM test was 6.13±1.06 (mean±S.E.) for the immunized group and0.96±0.90 for the control. The dif-ference between the two groups isstatistically significant, indicating thatGBP can effect cellular immunity inguinea-pigs. I-RNA and N-RNA were ex-tracted from spleens and lymphonodesof guinea-pigs immunized with GBPand of control animals respectively, andthen incubated in vitro with lympho-cytes isolated from 49 healthy persons.The lymphocytes treated with I-RNAreleased a factor which produced 7.73%slowing effect in the MEM test against GBP, while the lymphocytes incubatedwith N-RNA had only 1.29% slowingeffect. Statistical analysis of thesefigures showed a highly significant dif-ference between the two groups. Theseresults seems to suggest that I-RNAextracted from animals immunized withGBP can transfer immune reactivityagainst GBP from animal to humanlymphocytes.
