琼脂糖凝胶电泳及其应用于人血清β脂蛋白的分离和分型
AGAROSE-GEL ELECTROPHORESIS AND ITS APPLICATION TO THE SEPARATION AND CLASSIFICATION OF HUMAN SERUM LIPOPROTEIN
【摘要】 1.本文用自制琼脂糖进行人血清蛋白质和脂蛋白电泳及免疫电泳均获得较理想的结果。2.用酒精处理透析膜做为琼脂糖凝胶的载膜透明无色不吸附油红“O”染料,获得了较理想的载膜,便于长期保存和分析,极适宜于光密度描绘和测定各种脂蛋白组分和百分含量。3.在清蛋白的存在下以自制琼脂糖进行电泳获得了清晰透明整齐,分辨力强的电泳图谱,可将人血清脂蛋白分为五种主要的脂蛋白,乳糜微粒,β脂蛋白,前β2脂蛋白,前β1脂蛋白和α脂蛋白五种,以及两种不经常出现的脂蛋白,极慢速的前β3脂蛋白和前α脂蛋白。4.根据前β脂蛋白的出现与否,泳动的速度,出现的多少,染色的深浅以及乳糜微粒存在与否可将高脂蛋白血清电泳图谱分为八种不同的类型:即Ⅰ,Ⅱ1,Ⅱ2,Ⅱ3,Ⅱ4,Ⅲ,Ⅳ,及Ⅴ八种。5.自制琼脂糖电泳重复性较好,光密度测定同一标本获得了满意的结果。
【Abstract】 A simple method of preparing agarose gel suitable for immuno-electrophoresis and serum lipoprotein electrophoretic separation has been developed.By using a dialyzing membrane treated with alcohol as the supporting material for agarose gel electrophoresis a colorless transparent backgroud has been obtained. This transparent agarose gel eleotrophoretogram is excellent for optical scanning, thus the percentage of each lipoprotein component in the electrophoretogram can easily be measured.With this self-prepared agarose gel it is possible to separate serum lipoproteins in the presence of albumin into five clearly cut bands. The five major bands are chylomicrons, β-lipoprotein, pre-β2-lipoprotein, pre-β1-lipoprotein and α-lipoprotein. Besides, there have appeared infrequently two other fractions, a very slowly moving pre-β3-lipoprotein and pre-α-lipoprotein. It is, therefore, possible to classify the lipoprotein electrophoretograms according to the number and the rate of mobilities of the pre-β-lipoprotein, the intensity of the color of each band and the presence of chylomicrons into eight types, namely, Ⅰ, Ⅱ1, Ⅱ2, Ⅱ3, Ⅱ4, Ⅲ, Ⅳ, Ⅴ, each with its distinctive characteristics.The densitometry scanning determination of agarose gel lipoprotein electrophore-togram has given good results and satisfactory repetitions, so that a quantitative determination of each lipoprotein component can very easily be made.
