由生产胰岛素后的猪胰残渣和废液中用亲和层析法制备胰蛋白酶抑制剂

【摘要】 1.介绍了两种新的固定化胰蛋白酶的制备方法。一种是以微球型硅胶为载体,以戊二醛为交联剂,将猪胰蛋白酶吸附和固定在载体上。另一种是以甘蔗渣纤维素为载体,以对-β-硫酸酯乙砜基苯胺为双功能试剂,将猪胰蛋白酶共价偶联在载体上。两者都已成功地用作亲和层析的吸附剂,从工业生产胰岛素后的胰渣和废液中回收胰蛋白酶抑制剂。 2.提出了应用固定化胰蛋白酶的亲和层析技术从胰渣和废液中回收胰蛋白酶的工艺流程。经过中间试验,证明采用所制订的工艺流程,可以得到相当数量、具有一定纯度的胰蛋白酶抑制剂。 3.根据对所得抑制剂的初步分析,推测它是一种小分子量的硷性多肽,类似Kazal抑制剂。

【Abstract】 Either the waste pancreas residue subsequent to insulin extraction or the pH 6.0 mother liquor after crystallization of zinc-insulin can be used for the isolation of pancreatic trypsin inhibitor. An efficient method for separating and purifying the inhibitor which employs immobilized trypsin cross-linked on silica gel or covalently-linked to cellulose has been developed and tested on pilot-plant production scale.From the waste pancreas residue corresponding to 100 kg of fresh porcine pancreas, a total of 4.12 g of lyophilized powder of pancreatic trypsin inhibitor could be obtained, with a potency of 175-210 T. I. U. per mg lyophilized powder. An additional amount of 0.6 g of the inhibitor could be obtained from the pH 6.0 mother liquor, with a potency of 200 T. I. U. per mg lyophilized powder.A preliminary study of the properties of the porcine pancreatic trypsin inhibitor shows that the inhibitor is a basic polypeptide of relatively low molecular weight. It is different from the Kunitz inhibitor and resembles the Kazal inhibitor in inhi-.bition specificity.
