复健片促进MCAO大鼠中枢神经再生的机制研究

【摘要】 目的:探讨滋补肝肾中药复健片促进脑梗死后中枢神经再生的作用机制。方法:通过梳理古今对中风病的认识,结合导师多年的临床经验,提出肝肾阴虚是脑梗死的基本病机,观察导师验方复健片在大鼠MCAO后不同时相对BDNF、trk-B、Nogo-A、Omgp、NgR、slits、nestin、Brdu、NCAM、MAP-2、GAP-43及SYN等的变化,探讨复健片对神经再生营养因子、抑制因子、导向因子和内源性神经干细胞的增殖、迁移、分化以及轴突延伸和形成的影响。结果:复健片可增加神经生长营养因子表达,减少抑制因子表达,对神经元增殖、迁移、分化,轴突延伸、突触形成等有明显促进作用。结论:肝肾阴虚是脑梗死的基本病机,贯穿于脑梗死的始终。复健片可以通过多个靶点促进神经再生。

【Abstract】 Objective:To investigate the mechanism of Fujian Tablet on central nerve regeneration after cerebral infarction. Methods:On the base of analyzed the previous and modern documents, combined the abundance of clinical experiences of my tutor, we regarded the hepatic and renal yin deficiency as the basic pathogenesis of cerebral infarction. The effects of Fujian Tablet on the nerve regeneration nutrition factor, inhibitive factor, targeting factor and proliferation, migration, differentiation of endogenous nerver stem cell, axon elongation and form through observing the changes of BDNF, trk-B, Nogo-A, Omgp, NgR, slits, nestin, Brdu, NCAM, MAP-2, GAP-43 and SYN by hybridization in situ and immunohistochemical methods after Fujian Tablets interfered in the MCAO rats at different times. Results:Fujian Tablet increased the expression of neurotropic factor and targeting factor, decreased that of inhibitory factor. It promoted proliferation, migration, differentiation of neural stem cells, axonal growth and formation remarkably. Conclusions:Syndrome of hepatic and renal yin deficiency is the basic pathogenesis of cerebral infarction and resides in the course of cerebral infarction all the time. Fujian Tablet may promote central nerve regeneration by various of targets.
