中药新促孕液防治母畜子宫内膜炎的机理研究

【摘要】 子宫内膜炎是母畜常见的一种生殖系统疾病,严重影响着畜牧业的发展。西兽医常用抗生素和激素治疗,这些疗法虽有不同程度的效果,但西药的毒副作用和对食品安全的影响已引起人们的广泛关注,而用中药防治该病则可避免上述弊端。本课题组依据中兽医学理论研制成功中药新促孕液(NFPL),用于防治母畜子宫内膜炎取得了可靠疗效。为进一步改进组方、提高疗效,本研究系统观察了NFPL及其组分药、相关单味药和其加味方对子宫内膜炎主要病原菌生长、母畜血液流变、子宫内膜结构和子宫运动性等方面的影响,从清热燥湿、活血化瘀和祛腐排脓等方面探讨了NFPL的作用机理。 试验Ⅰ 奶牛子宫内膜炎主要病原菌的分离及鉴定 从南京地区奶牛场采集临床型子宫内膜炎患牛子宫分泌物进行病原菌分离及鉴定,为研究NFPL的抑菌机理和制备疾病模型准备材料。结果,共分离出葡萄球菌10株、链球菌6株、大肠杆菌8株,溶血性和非溶血性大肠杆菌的血清型分别为O159和O110,O159的致病性较强,可在68h内引起小鼠全部死亡。表明葡萄球菌、链球菌和大肠杆菌是引起奶牛子宫内膜炎的主要病原菌。 试验Ⅱ NFPL及其组分药和相关单昧药的抑菌作用测定 为了研究NFPL的清热燥湿作用和改进组方,测定了NFPL及其组分药和其他10味中药对子宫内膜炎病原菌主要分离菌株的体外抑菌圈直径和最小抑菌浓度。结果表明,NFPL对6个分离菌株均具有较强的抑制作用;NFPL的组分药以淫羊藿、黄柏和硼砂的抑菌作用较强,其他单味药以丹参的作用最强,可以作为NFPL改进方的组分药。 试验Ⅲ NFPL及其主药和相关单味药影响血液流变的作用测定 为了观察NFPL的活血化瘀作用和改进组方,选用25只健康家兔随机分为5组,4个试验组分别用NFPL、红花、丹参和川芎煎剂灌胃,对照组灌给等量生理盐水,每天1次,连续7d,末次给药后耳缘静脉注射高分子葡聚糖致瘀;分别于首次给药前、用药后和致瘀后1h采血,用MVIS-2035型全自动血液流变分析系统测定血液流变学参数的变化。结果表明,NFPL能显著降低健康家兔的血液粘度,并能显著抵抗高分子葡聚糖的致瘀作用,其

【Abstract】 Endometritis is one of common reproductive diseases in female animals, badly impacting the development of animal husbandry. Antibiotics and hormones have been the first priority for the prevention and treatment of this disease. However, the toxicity and side-effect of this therapy for food safety has been paid more and more attention although the therapy still works to some extent clinically. To avoid the toxicity and side-effect of these antibiotics and hormones used in veterinary clinic, the Chinese herbal medicine were developed for the purpose of the prevention and treatment of dam endometritis. Based on the traditional Chinese medicine theory, our research group successfully developed a Chinese herbal medicament known as New Fertility-promoting Intrautenne Infusion Liquid (NFPL) with definite and stable curative effects on endometritis. In order to further improve prescription and curative effects of this drug, the effects of NFPL and its component herbs in comparison with other Chinese herbs and additive prescriptions on growth of pathogenic bacteria caused endometritis, hemorheological parameters, endometrium structure and uterus motility were determined, and its mechanism was studied from the aspects of clearing away fever and drying dampness, activating blood flow and removing blood stasis, expelling putrefaction and eliminating pus in the present study.Experiment I Separation and identification of pathogenic bacteria caused endometritis in dairy cattle The experiment was designed for the preparation of pathogenic bacteria used for the detection of antibacterial activity with Chinese herbal medicament - NFPL and its component herbs, as well as for the establishment of pathological model of rabbit endometritis. The uterine secretion from dairy cow with clinical endometritis in Nanjing area was sampled and the pathogenic bacteria were separated and identified as routine protocol, resulting in 10 strains of Staphylococci, 6 strains of Streptococci and 8 strains of E. coli obtained.
