块型重组合异种骨的研制及其相关基础研究

【摘要】 骨移植术是骨科治疗骨缺损、骨不连最常采用且最有效的方法之一,在应用数量上骨移植已经成为仅次于输血的人体组织移植。自体骨移植虽然成骨活性好,不存在免疫排斥反应,但不可避免地造成供骨区的医源性破坏,增加患者痛苦,并且供骨量十分有限。因此,研制自体骨移植的替代材料,一直是骨科及相关学科长期以来的热门课题。异种骨取材广泛,价格低廉,便于储存,但异种骨移植因发生剧烈的免疫排斥反应,最终导致失败。本论文在颗粒性重组合异种骨研究的基础上,研制出块型重组合异种骨(massive reconstituted bone xenograft,MRBX),并针对异种骨移植中存在的问题进行了系列基础研究。 一、异种松质骨移植抗原分布的免疫组化研究 利用经二次免疫BALB/c小鼠获得的小鼠抗牛松质骨多克隆抗体血清,采用双PAP法及免疫荧光法对新鲜牛松质骨脱钙石蜡切片进行了免疫酶及免疫

【Abstract】 Xenogeneic bone grafting is an alternative to autogeneic bone grafting, but the intense immune rejection makes its clinical application impossible . The experimental studies in the present report are designed to address this problem.1. Immunohistochemical Location of Xenograft Antigens in Bovine Cancellous BoneMouse anti-bovine cancellous bone polyclonal antibody (MABCB PcAbs) was produced in 8 BALB/c mice immunized twice with supernatant fluid of homogenate of fresh bovine cancellous bone. Immunohistochemical study was performed to locate the antigens of bovine cancellous bone xenograft with MABCB PcAbs by both double PAP method and immunofluorescence technique. In bovine cancellous bones, a strong reactivity was found in blood cells, bone marrow cells, osteocyte, osteoblast, chondrocyte, chondroblast plasma and osseous lacuna, but no immunostaining was detected in bone matrix.
