大鼠膀胱kit阳性间质细胞在逼尿肌自发性收缩发生机制中作用的相关研究

【摘要】 【目的】 1.探讨大鼠kit阳性间质细胞在膀胱壁内的分布、超微结构特征,与平滑肌、神经之间的微解剖关系。 2.研究大鼠膀胱逼尿肌自发性收缩活动的一般特征,探讨膀胱kit阳性间质细胞在其发生机制中的作用。 3.探讨kit阳性间质细胞在不稳定膀胱发病机制中的作用,以及kit受体酪氨酸激酶对这一病理过程的影响,为不稳定膀胱的治疗提供新的途径。 【方法】 1.我们运用免疫组化的方法来确定kit阳性间质细胞在膀胱壁内的组织学分布。通过透射电镜观察其超微结构特征、与其他细胞之间的缝隙连接结构以及与神经节、神经纤维束间的位置关系。同时运用扫描电镜观察这种细胞形态特征。 2.通过张力换能器和描记仪记录组织浴液中离体逼尿肌组织条自发性收缩活动,观察逼尿肌自发性收缩活动对多种离子通道阻断剂、神经递质、受体阻断剂以及kit受体特异性阻断剂Gleevec的反应。对离体逼尿肌组织进行亚甲基兰染色,观察光照前后逼尿肌自发性收
【Abstract】 Objects:to identify the presence and distribution of kit positive interstitial cells in rat bladder smooth muscle and characterize their morphology and ultrastnictural features;to study whether the kit positive interstitial cells played a role in the mechanisms generating spontaneous activity events;to evaluate the relation between kit receptor tyrosine kinase and the physiology function of kit positive interstitial cells and alterations of kit receptor tyrosine kinase at conditions of excessive mechanical overload.Materials and methods:Bladder obtained from the normal and mode of detrusor overactivity in rat were prepared for immunohistochemical investigations using the primary antibody of the kit receptor tyrosine kinase, and ultrastnictural studies using transmission electron microscope. Enzymatically dispersed cells were showed by scanning electron microsope. In this study,we had characterized the spontaneous activity of the urinary bladder smooth muscle from normal rat, the mode of detrusor overactivity induced partial outlet obstruction and stained by methylene blue,partically MB plus light using isometric muscle tension recording techniques.We also examined effets of Gleevec on spontaneous activity ,and accessed mRNA of kit receptor tyrosine kinase at the conditions of excessive mechanical overload through RT-PCR.
