白色念珠菌对口腔角质形成细胞移行能力及其相关信息通路影响的体外研究

【摘要】 研究背景 近年研究发现,细胞移行(cell migration)不仅在胚胎发育、免疫反应、创伤修复和组织再生等生理过程中发挥着重要作用,且参与慢性炎症、血管疾病和肿瘤转移等病理过程。白色念珠菌(candida albicans,C.albicans)是机体中最常见的条件致病菌,C.albicans对宿主细胞的粘附以及在此过程中C.albicans与宿主细胞的交互作用亦成为近年广受关注的研究课题。我们推论:C.albicans感染宿主细胞后可通过细胞内信息流改变宿主细胞的移行功能,但缺乏直接的研究证据证实。本课题以口腔黏膜角质形成细胞株为研究对象,用C.albicans刺激细胞后观察细胞移行功能的变化以及与移行密切相关的细胞外信号调节蛋白激酶(extracellular signal-regulated protein kinase cascade,ERK)和灶性粘附激酶(focal adhesion kinase,FAK)信息通路的变化,来探索C.albicans与口腔黏膜角质形成细胞交互作用过程中是否会改变细胞的移行能力,并对其相关信息通路进行研究。 研究目 的在体外观察C.albicans感染口腔黏膜上皮细胞后是否诱导细胞移行发生改变,并初步探讨与细胞移行密切相关的ERK和FAK通路的变化情况。 研究方法 以口腔角质形成细胞株Tca8113作为研究对象,①用western

【Abstract】 Background: The importance of cell migration is evident from both the physiological processes that depend on the regulated movement of cells, including embryonic development, immune responses and tissue maintenance and repair, and from the disease states driven by aberrant cell motility, such as chronic inflammation, vascular disease and tumor metastasis. Not surprisingly, immense effort has been directed towards furthering our understanding of this complex process. Extracellular signal-regulated protein kinase cascade (ERK) is one of best characterized intracellular signaling pathways, which play a critical role in the control of cell survival, proliferation, and migration when activated by series of stimuli. Recently available information suggests that Candida albicans (C.albicans) have relationship with cell migration.
