整合子在铜绿假单胞菌中的分布及其在生物被膜中作用机制的研究

【摘要】 铜绿假单胞菌是临床常见的条件致病菌,耐药性突出。新近研究发现该菌在感染过程中常以生物被膜形式存在,整合子在其耐药性形成和播散中起重要作用。本研究的目的是探讨整合子在我国铜绿假单胞菌临床分离株中的分布,以及整合子在该菌生物被膜中的作用机制。 本研究采用多重聚合酶链反应技术对分离自河北和昆明两地的82株铜绿假单胞菌临床分离株进行Ⅰ类、Ⅱ类和Ⅲ类整合子的检测,结果51株(62.2%)为Ⅰ类整合子阳性菌,1株(1.2%)为Ⅱ类整合子阳性菌,未检出Ⅲ类整合子阳性菌。其中Ⅱ类整合子在铜绿假单胞菌中的存在未见报道。采用聚合酶链反应技术,在51株Ⅰ类整合子阳性菌中检测到28株(54.9%)携带Ⅰ类整合子相关基因盒。药敏结果显示Ⅰ类整合子基因盒阳性菌株对哌拉西林、头孢哌酮、头孢他啶、妥布霉素、阿米卡星、环丙沙星、氯霉素和复方磺胺甲嗯唑等抗生素的耐药率显著高于未携带整合子相关基因盒的菌株(P<0.01),提示Ⅰ类整合子基因盒在铜绿假单胞菌临床分离株中发挥耐药作用。Ⅰ类整合子在石家庄、昆明两地铜绿

【Abstract】 Pseudomonas aeruginosa is an important opportunistic pathogen of human with high drug resistance. Recent researches have revealed P. aeruginosa readily forms biofilm, which displays high resistance to antibiotics. And most of P. aeruginosa clinical isolates tested carried integrons, which played important roles in the spread of antibiotic resistance gene and multidrug resistance. The aim of this study are to detecte the distribution of integrons among P. aeruginosa clinical strains and to approach the mechanism of integron in their biofilm.In this study, the distribution and characterization of the class 1、2 and 3 integrons, among 82 P. aeruginosa clinical strains from Shijiazhuang and Kunming, were detected with specific primers of class 1 integrase gene (intI1), intI2 and intI3 by multiplex- PCR. In the result, 51 (62.2%) were positive for intI1, one was positive for intI2 and no one positive intI3 among these strains. Among 51 intI1 -positive strains, 28 (54.9%) were amplified by primers of in-F and in-B which were set for amplifying the region of resistance gene cassettes.
