抑郁舒抗抑郁作用和相关功效研究

【摘要】 抑郁状态是由各种原因引起的心境障碍(mood disorders)或情感性障碍(affective disorders),是以抑郁心境为中心的临床症状群或状态。常见于情感性精神障碍的抑郁症(单相抑郁症)、抑郁性精神症、焦虑性精神症、反应性抑郁症等,并可伴发于其他疾病中。有关抑郁状态的疾病,尤其是抑郁症,其发病率高,不仅给患者自身健康及其家庭生活带来困扰,而且也给社会、国家带来重大负担,已引起国际社会的关注。近年来,中医针对抑郁症或抑郁状态进行辨证论治,取得了一定的临床疗效,且不良反应少,而受到医药界的重视。但目前处方用药比较分散,较难推广。 本课题对抑郁状态的中医药认识、治疗及相关方药的研究现况作了较广泛性的文献探讨,并采用小鼠悬尾模型,大、小鼠迫游泳模型,利血平拮抗模型等多种抑郁模型,观测了抑郁舒与香附对各抑郁模型的药效指标的影响,并对二者的抗疲劳、镇静安眠等作用进行了实验研究。结果表明: ①与空白对照组相比较,抑郁舒能显著改善小鼠迫泳模型、小鼠悬尾模型及利血平拮抗所致的抑郁模型相应指标。同时,香附对小鼠悬尾模型、大鼠迫泳模型亦有一定改善作用。结果表明抑郁舒具有对抗抑郁状态的作用。 ②抑郁舒能明显提高利血平导致的抑郁状态小鼠模型动物脑内的去甲肾上腺素(NA)、多巴胺(DA)和五羟色胺(5-HT)含量。表明抑郁舒通过提升大脑中单胺神经递质含量从而改善抑郁状态。 ③抑郁舒能有效地延长小鼠负重游泳时间及提升红细胞中超氧化物歧化酶(SOD)活性,表明抑郁舒能改善抑郁状态造成的疲劳。 ④抑郁舒与香附都能减少小鼠的自主活动量,表明抑郁舒与香附都具有一定的镇静作用。同时,二者都能延长受戊巴比妥钠影响下小鼠的睡眠时间,表明抑郁舒与香附都能改善抑郁状态造成的睡眠障碍。

【Abstract】 Depressive syndrome is a group of pathological conditions characterized by in common by the depressive mood state .Diseases as mood disorders (depression, mania) and other psychiatric disorders are included, and could be presented in other pathologies. It’s a syndrome of high incidence, difficult to treat, and will cause great loads to the helth of patients and their families. Tt has causes a great impact among the world. Chinese traditional medicine makes great results to ameliorate this syndrome with little side effects in recent years. But, the choosing criteria of correct formula to treat depressive syndrome are dispersive for now. So, it is difficult to popularize that.In this study, Yiyushu and Rhizoma Cyperis are compared for their possible antidepressant effect in animal models as: tail- suspension model of mice, forced swimming models of mice and rat, and reserpine induced depression model in mice, as well as their anti-fatigue, depressant activity of central nervous system and hypnotic activity. Results of the research can probe:1 Yiyushu shows important effects compared with control group in forced swimming model of mice, tail- suspension model of mice and reserpine induced depression model. In the same time, Rhizoma Cyperis shows significant effects in tail- suspension model of mice and forced swimming model of rats. The above results suggest that Yiyushu could have significant effect for treatment of depressive syndrome.2 Yiyushu clearly elevates concentrations of noradrenalin (NA), dopamine (DA) and serotonin (5-HT) of brain in mice under depression status caused by reserpine, suggests that Yiyushu could improve depressive status through elevations of the monoamine neurotransmitters.3 Yiyushu evidently prolongs the swimming time and superoxide dismutase
