Urocortin抗大鼠心肌缺血—再灌注损伤及高血压心脏重构机制的研究

【摘要】 背景 优洛可定(urocortin)是40个氨基酸的小分子肽,是CRF肽类家族的一个新成员,参与哺乳动物应激反应的一种关键因子,除下丘脑外,它在中枢其它部位和外周组织中也发挥多方面作用。CRF肽类家族主要通过与相应G-蛋白偶联的CRF受体(CRF-R1,CRF-R2)相结合,调节机体内分泌、自律性以及对应急的行为反应等。Urocortin最早在中枢神经系统内发现,近年越来越多的研究表明urocortin可以通过自分泌和/或旁分泌方式与CRF受体相结合在心血管系统发挥重要作用,并且它的这种心血管作用比CRF更显著。有报道表明,urocortin能显著而持久的降低动脉血压,增加动脉血流量,升高心率,增加冠脉输出。在体及离体实验研究发现,在缺血-再灌注损伤、动脉粥样硬化以及高血压等心血管病症中,urocortin能保护心肌细胞免受损伤,起到心血管保护作用。Urocortin预处理可增强心肌抵抗力,其中最重要的途径很可能是:urocortin与CRF-R2结合后直接调节PI-3K,经过Ras、Raf-1激活MEK1/2、ERK1/2-P42/44 MAPK、PKC/KATP通道,后者通过磷酸化激活多种转录因子及降低氧自由基生成等一些现在尚不十分明确的途径,发挥心血管保护作用。本实验室在体实验研究结果表明,urocortin能显著降低缺血-再灌注对大鼠心肌损伤。另外,实验中发现urocortin能抑制血管平滑肌细胞增殖,这也提示urocortin有可能可以逆转心室重构,在高血压等疾病中起心血管保护作用。 目的 1.研究urocortin通过降低氧化应激反应对缺血-再灌注损伤大鼠模型的保护作用机制; 2.通过研究urocortin对缺血-再灌注损伤大鼠模型超微结构改变的影响,进一步阐述其在缺血-再灌注损伤中的保护作用; 3.探讨长期及亚急性应用urocortin对自发性高血压大鼠血流动力学

【Abstract】 BACKGROUNDUrocortin (UCN) is a 40-amino-acid peptide related to the hypothalamic hormone corticotrophin releasing factor (CRF), the central mediator of the hypothalamic-pituitary-adrenal axis and stress response in mammals. CRF family exert their physiological effects through binding to two G protein-coupled receptors (CRFR) 1 and 2, to modulate endocrine, autonomic, and behavioral responses to stress. Urocortin was firstly found in the central nervous system. However, recent studies showed that it has much more potent effects on the cardiovascular system than CRF. It enhanced cardiac contractility, coronary blood flow, heart rates and cardiac output. Some in vitro & ex vivo investigations showed that in case of ischemia/reperfusion (I/R), atherosclerosis and hypertension, urocortin could protect cardiac cells from severe injury. Pretreatment of cardiac myocytes with urocortin could improve myocytes’ condition via PI-3K, Ras, Raf-1 passway to active the mitogen-activated protein kinase (MAPK) pathway, protein kinase C and ATP sensitive K+ (PKC/KATP) channels etc. Our recent in vivo study demonstrated that urocortin could reduce cardiac damage in I/R model. Moreover, we firstly found that urocortin could inhibit the proliferation of the vascular smooth muscle cells, highly suggesting that urocortin might play a role in reversing cardiovascular remodeling and urocortin has the potent protective effects in cardiovascular system. AIM 1. The aim of this study was to investigate the effects of urocortin onoxidative stress and the mechanisms of urocortin on ischemiareperfusion (I/R) injury in vivo in the rat model.
