低强度压应力对于体外培养的软骨细胞代谢的影响

【摘要】 骨关节炎的发病人数及其所带来的危害要明显超出我们的认知范围。在最常见的三大老年疾病中,骨关节炎的发病率及患病人数在世界范围内均位居首位。 关节软骨的代谢特征及细胞因子在关节软骨代谢中的影响意义正成为骨关节炎发病机理研究的一个焦点。 应力在骨关节炎形成及发展中的参与机制尚不明确。细胞因子的来源及影响细胞因子产生的方式并没有进行充分的研究。尤其应力因素与细胞因子产生的确切关系尚未明确。 在软骨组织修复的治疗方法中,自体软骨细胞移植正成为目前研究和应用的重要方法。然而,如果不建造与体内物理(如应力环境)及生化环境相类似的体外培养条件,同时忽略软骨细胞移植后所处的应力及生化环境对其自身可能的影响,自体软骨细胞移植这一有希望的治疗方法将很难最终得以完善。 综合骨关节炎研究的现状及治疗上存在的问题,不难发现其内在的一个共同原因,那就是:目前我们还缺乏在细胞、分子层次上对于软骨组织、软骨细胞与各种骨关节炎致病因素之间应答特点的充分理解。 本实验选择压应力作为影响软骨细胞代谢功能的外加条件,目的在于: 1.观察不同压应力强度、压应力作用时间及间歇、持续两种压应力形式对于软骨基质代谢、软骨细胞增殖的影响。 2.观察两种形式的压应力对于软骨细胞产生细胞因子的影响。 3.探索对于软骨细胞代谢构成负性影响的应力环境,在此基础上建立特定压应力与软骨细胞代谢抑制、软骨组织损伤因子的体外关系。然后,观察药物及各种调整性的应力对于这种关系的影响。 4.探讨软骨细胞体外培养适宜的力学条件。 实验工作结果显示: 1.低强度压应力能够明显影响软骨细胞的代谢、增殖活动,在不同的低强度压应力环境下具有丰富的反应特征。在压应力作用的后期,软骨细胞的代谢、增殖活动呈抑制趋势。
【Abstract】 Osteoarthritis (OA) is one of the most common and severely harmful diseases, which is more significant than our recognition. Currently, its morbidity and the number of its victims are both the highest among the three most common diseases in older people throughout the world.The characteristic of cartilaginous metabolism and the effect of cytokines in cartilaginous metabolism are becoming one of the research focuses of OA study.However, the mechanism of mechanical forces in OA occurrence and development is poorly understood. The origin of cytokines and the way stimulating cytokines production, especially the exact relationship between mechanical forces and cytokine production are not studied sufficiently.In order to repair chondral defects, autogenous chondrocyte transplantation is becoming encouraging treatment concerned by most investigators. However, this method will not succeed ultimately if no suitable cell culture environment in vitro can be found just as the environment in vivo both in biomechanical condition(such as stress system) and a biochemical condition is constructed and if we overlook the potential effect of mechanical and biochemical condition on autogenous grafts after transplantation.When reviewing the problems existing in current study and treatment of OA, it is easy to find that lacking sufficient understanding the relationship between chondrocytes and different causes of OA on cellular and molecular levels is the most important difficult.It is for four purposes that we induced compressive stress as an effect on chondrocytes metabolism in this study. First is trying to clarify the effect of compressive stress applied in different ways such as different strength, different acting time and different frequency on chondrocytes matrix metabolism and proliferation. Second is to define the effect two types of compressive stress, continuous and intermittent, on cytokine produced by chondrocytes. Third is to explore the stress condition which have a reversed effect on chondrocyte metabolism for expliciting the relationship between
