电磁辐射净化自体造血干细胞移植物中残留白血病细胞的机制研究

【摘要】 造血干细胞移植是血液学领域一项划时代的新技术,是治疗恶性血液肿瘤、实体瘤以及某些遗传性疾病的重要方法。目前,对于白血病人的造血干细胞移植,在动员外周造血干、血细胞分离机采集干细胞后一般没有采取进一步的净化措施。如何进一步有效净化血液病人自体造血干移植物中残留的白血病细胞成为延长病人无病生存期的关键技术。本文利用分子生物学、细胞生物学手段,研究电磁场对白血病细胞的净化效应及其机制,探讨毫米波应用于白血病人自体造血干移植物体外净化的可行性。本文共七章,各章主要研究内容及结果如下: 第一章,综述了自体造血干细胞移植的进展及电磁场生物效应研究领域的新成果,及国内外利用电磁场诱导肿瘤细胞凋亡的研究结果及其机制。 第二章,利用电镜技术、DNA末端标记、MTT活力测定等方法,研究了毫米波对红白血病K562细胞株、人类急性早幼粒白血病细胞株HL-60及正常人骨髓单核细胞的细胞超微结构、细胞凋亡、细胞活力的影响,结果发现毫米波辐射对正常人骨髓细胞及白血病细胞的影响明显不同,正常人骨髓细胞所受影响较小,特别是功率密度小的毫米波对其无影响;而白血病细胞受辐射后发生显著的凋亡或坏死,细胞活力显著下降,该效应具有明显的功率窗和时间窗。 第三章,利用流式细胞术定量研究了毫米波辐射白血病细胞后的细胞内钙离子浓度变化及NO的代谢规律,结果表明毫米波辐射可通过影响细胞内钙离子浓度及NO的代谢进而损伤K562细胞,细胞内这两种信号系统在毫米波辐射杀伤白血病细胞过程中都发挥重要的作用。 第四章,研究了毫米波辐射对HL-60细胞内自由基及抗自由基酶系的影响,发现毫米波辐射影响自由基的瞬时浓度,并进而产生一系列过氧化效应,同时对细胞内重要的抗氧化酶系的活性有显著的抑制作用,该作用表现出一定的频率相关性和时间相关性。 第五章,采用特异性抗体结合流式细胞术研究了毫米波辐射对K562白血病细胞株bc1-2基因表达的影响,发现在毫米波辐射24h后,K562细胞的bc1-2基因表达受到明显抑制,bc1-2基因是毫米波辐射后调控K562细胞生理代谢变化的后期因子。

【Abstract】 Autologus hemetopoietic stem cell transplantation is an important new technologic method to cure malignant blood tumor , substance tumour as well as some heredity diseases . At present, as for the autologus hemetopoietic stem cell transplantation for leukemia, further purification was not adopted after blood cells were collected. How to make the stem cells purified effectively, becomes a key technology to lengthen survival time of patients. In these studies, molecular biology and cytobiology methods were used to research the mechanisms of the purification effect of electromagnetic irradiation.There are seven chapters in these studies, contents of each chapter are as below:In Chapter One, the progresses of autologus hemetopoietic stem cell transplantation researches and electromagnetic biological effect researches were summarized;In Chapter Two, some methods such as electron microscope, DNA ending label and MTT measurement were used to studied the changes of apoptosis , necroses and cell vigour of leukaemia K562 cell trunk and HL-60 cell trunk irradiated with millimeter wave. Different effects of millimeter wave to leukaemia cell trunks and human marrow cells were observed clearly. Human marrow cells were not so sensitive to millimeter wave radiation as leukaemia cells.In Chapter Three, the FCA method was used to studied the changes of calcium ion consistency and the content of NO inside the leukaemia cells after irradiated with millimeter wave. It was found that millimeter wave radiation may harm leukemia cells by regulating calcium ion consistency and NO inside the cells.In Chapter Four, the changes of free radical and antioxidase in HL-60 cells after irradiated with millimeter wave were studied.
