三七复方成分对人子宫内膜细胞炎症模型的影响

【摘要】 炎症因素所致的子宫异常出血是妇产科临床常见病症,如急慢性盆腔炎、子宫内膜炎、宫内节育器、宫内节育器或药流人流后出血、产后出血等均与炎症有直接或间接关系。传统中医药对妇产科“血证”治疗效果显著、颇具特色。三七是化瘀止血名药,在妇产科“血证”中尤为常用,但其具体的作用途径仍不清楚。本课题建立了由细菌脂多糖诱导的人子宫内膜细胞的炎症模型,并对炎症细胞模型施加三七复方成分,通过观察子宫内膜局部炎性细胞因子及血管活性物质的变化,来探讨三七复方成分治疗炎症性子宫异常出血的作用机制。结果显示: 1.通过MTT法观察三七复方成分作用于人子宫内膜细胞的细胞毒作用,确定了三七复方成分干预子宫内膜细胞的最佳浓度(三七皂苷0.0408mg/ml+三七氨酸0.002mg/ml)。 2.三七复方成分、三七皂苷及三七氨酸都能有效的抑制人子宫内膜细胞培养液中炎性细胞因子白细胞介素1β和肿瘤坏死因子-α的分泌,但在降低白细胞介素1β和肿瘤坏死因子-α浓度的幅度上,三七复方成分明显强于三七皂苷及三七氨酸。 3.三七复方成分、三七氨酸能明显降低炎性子宫内膜细胞培养液中一氧化氮、一氧化氮合酶及纤溶活性,调整6-K-PGF1α/TXB2比值,使之趋于正常。而三七皂苷在短时间内作用与三七复方成分及三七氨酸相反。 上述实验结果表明:三七皂苷和三七氨酸两者互相协同,通过抑制子宫内膜局部炎性细胞因子的释放及及对血管活性物质的调整,从而实现三七复方成分对炎症性子宫异常出血的良好防治作用。
【Abstract】 The abnormal bleeding of uterus such as acute or chronic pelvic inflammatory disease , endometritis, bleeding after Cu-IUD-Wearing , medical abortion or induced abortion , and flooding are the most popular female diseases. These diseases are directly or indirectly relative to inflammation. Panax Notoginseng, a famous traditional Chinese medicines, has significant effect on the treatment on these abnormal bleeding. However its working mechanism of removing blood stasis, hemostasis and anti-inflammation is unclear so far. In this thesis work, the cell model of inflammatory human endometrium was built up and intervented with compound components of panax notoginseng. And the working mechanism of compound components of panax notoginseng on the abnormal bleeding was explored by the change of cytokine andvaso-active factor of endometrium. The results show:1. the most proper concentration of compound components of panaxnotoginseng (panax notoginseng saponines 0.0408mg/ml and dencichen 0.002 mg/ml) was determined by the MTT assay of compound components of panax notoginseng on the cells of endometrium.2. The compound components of panax notoginseng , panax notoginseng saponines and dencichen all could inhibit the secretion of cytokine effectively such as TNF-aand IL-1(3 in the RPMI1640 of human endometrial cell. But the compound components of panax notoginseng was the most powerful of all .3. The compound components of panax notoginseng and dencichen both could distinctly decrease NCK iNOS and fibrinolytic activity and adjust the odds of 6-K-PGFla / TXB2 of the RPMI1640 of human endometrial cell to near normality. But the effect of panax notoginseng saponines was opposed to the both in a short time.All mentioned above indicate that the effect of panax notoginseng saponines and dencichen on ihibiting the release of cytokine and adjusting vaso-active substance of endometrium is synergistic, So the compound components of panax notoginseng can prevent and cure the abnormal bleeding of uterus favorably caused by inflammation.
