乌药与鼎湖钓樟的化学成分及抗HIV-1整合酶活性研究

【摘要】 乌药(Lindera aggregata(Sims)Kosterm.)为樟科(Lauraceae)山胡椒属植物,《中国药典》(2000版)规定其块根入药,为传统的温中理气止痛药,具有抗菌、抗病毒、保肝等药理活性。文献报道乌药叶具有抗炎、抗菌等作用,现在药材市场上所售乌药块根饮片中多掺杂大量乌药茎饮片。鼎湖钓樟(Lindera chunii Merr.)为乌药的民间代用品,其根入药,又称为“台乌球”。乌药地上部分与鼎湖钓樟的化学成分研究尚未见报道。为开发利用乌药资源,本文对乌药与鼎湖钓樟进行了系统研究。 利用硅胶,聚酰胺,ODS,Sephadex LH-20等分离材料,从乌药与鼎湖钓樟中分离并鉴定44个化合物,其中6个为新化合物和1个为首次从天然界分离得到。结果如下: 从乌药叶中分离并鉴定18个化合物:去氢香樟内酯(1),strychnistenolide 6-0-acetates A and B(2A和2B),β-谷甾醇(3),乌药内酯(4),羟基香樟内酯(5),山奈酚(6),槲皮素(7),nubigenol(8),山奈酚-3-0-(6”-反式-对-肉桂酰基)-β-D-吡喃葡萄糖苷(9),香叶木素-7-0-β-D-吡喃葡萄糖苷(10),槲皮素-3-0-鼠李糖苷(11),芦丁(12),山奈酚-3-0-L-吡喃阿拉伯糖苷(13),胡萝卜苷(14),槲皮素-3-0-β-D-吡喃半乳糖苷(15),异鼠李素-3-0-[葡萄糖-(6-1)-鼠李糖苷](16),山奈酚-3-0-葡萄糖醛酸苷(17)。化合物8,9,10,16,17为属内首次分离得到。 从乌药茎中分离并鉴定10个鞣质类化合物,其中1个为新化合物:epicatechin(4β-8,2-0-7)-catechin(4β-8)-catechin(lindetannin trimer,26);9个为已知化合物:(+)-儿茶素(18),(-)-表培儿茶素(19),procyanidin A2 and A6(20 and 21),(-)-表儿茶素(22),procyanidin B1 and B2(23 and 24), cinnamtannin B1(25),epicatechin(4β-8)-epicatechin(4β-8,2-0-7)-epicatechin(4β-8)-epicatechin(27)。并对缩合鞣质类化合物的一些波谱学规律进行小结。 从鼎湖钓樟的块根中分离得到20个化合物,其中5个为新化合物:lindenanolide E(28)、lindenanolide F(29)、lindenanolide G(30)、lindechunine A(41)、lindechunine B(43);1个为首次从天然界分离得到:lindenanolide H(31);14个为已知化合物:strychnistenolide 6-0-acetates A and B(2A和2B),β-谷甾醇(3),乌药内酯沈阳药科大学博士学位论文(4),pseudoneoliderane(32)、乌药醇(33)、hem。dLne(34):N-heruangerine(35)、ocokl-yptlne(36)、hem。donine(37)、N-methylhe。angerine(38)、新木姜子碱(39)、7 oxohernaglne (4)、7 oxohernangerine(42)。 运用 RPWLC色谱法,建立了乌药块根与茎中缩合鞍质三聚体(化合物 25和 26)的含量测定方法,并采用皮粉法测定总斡质含量,茎的总跷质含量为3.sffe,块根的总辑质含量为5.43q0。 以HIV-l整合酶为靶点,采用MIA(MUltiPlate[ntegyation Assay)法,对乌药与鼎湖钓樟中分离得到的29个化合物进行活性筛选。结果表明,乌药中缩合摇质二、三、四聚体和鼎湖钓樟中4个阿朴菲型生物碱具有抑制HIV叫整合酶的活性:ProcyanidinB;(23,28.6帅,Procvanidin巳(24,31.3叫),cinnmt。洲in B;(25,& 3卜),oicatechin(4 P-8,2—0-7)-catechin(4 g-8)-catechin(26,7.gW),印catechin(4尸-8)-epicatechin(4尸一8,2-O-刀-epic。ltechin(4尸-幻-p icatechin(27,5.Zgh),hernandonine(37,16.3gb),新木姜J’碱(39,7.7po,lindechunine A*,ZI.l卜),7-oxohernanrerine(42,18.2刚。户阿朴菲型生物碱的构效关系进行初步探讨:连h氧亚甲基为7-氧化阿朴菲型生物碱的活性基团;对于非氧化阿朴菲型生物碱,C-11位无取代基的化合物有可能具有较强抑制活性。 最后,对乌药与鼎湖钓樟的资源分布及植物形态进行比较,两种植物的形态有显著区别:通过对乌药与鼎湖钓樟的块根和叶的显微结构的观察,发现鼎湖钓樟的块根中含有石细胞群。

【Abstract】 Lindera aggregata (Sims) Kosterm. is a plant of Lindera genus and used as a palliative and an antispasmodic drug. The Leave of L aggregata (Sims) Kosterm. has antibacterial, anti-inflammatory effects. The root of L chunii Merr. is frequently used as a substitute of Wuyao in some places of Guangdong provinces. However the absence of phytochemical investigation impedes further development and application of two plants, these inspired us to study their chemical constituents.Chemical investigation of two plants has resulted in the isolation of 44 compounds including 6 new compounds and 1 naturally occuring compound, their structures were identified on the basis of UV, IR, NMR, MS, physicochemical properties and literature data. The results were shown as follow:18 known compounds from leaves of L aggragtawere dehydrolindestrenolide (1), strychnistrenolide 6 - 0 - acetates A and B(2A and 2B), β - sitosterol(3), lineralactone(4), hydroxylindestrenolide(5), kaempferol (6), quercetin(7), nubigenol (8), kaempferol - 3- 0 - (6" - trans - P - coumaroyl) - 0- D - gluco pyranoside(9), chrysoeriol - 7 - 0 - β?D - glucopyranoside (10), quercetin -3 - 0 - rhamnoside(ll), rutin(12), kaempferol - 3 - 0 - L - arabinopyranoside (13), daucosterol(14), quercetin 3-0- β -galactopyanoside(15), isorhamnetin 3 - 0 - (glucopyranosyl - (6 -1) - rharanoside) (16) and kaempferol - 3 - 0 - β - glucuronoside(17). Compounds 8, 9, 10, 16 and 17 were firstly isolated from this genus.10 compounds were isolated from stems of L aggregata including 1 new compound : epicatechin - (4β - 8, 2-0 -7) - catechin (4 β- 8) - catechin (lindetannin trimer, 26), and 9 known compound : (+) - catechin (18), (-) -epigallocatechin(19), procyanidin A, and Ag (20 and 21), (-) - epicatechin (22), procyanidin B1 and B2 (23 and 24), cinnamtannin B, (25), epicatechin (4β - 8)- epicatechin (4β -8, 2-0-7) - epicatechin (4β - 8) - epicatechin (27).3 new eduesmane type sesquiterpenoids: lindenaiolides E(28), F(29) and G(30), and two new aporphine alkaloids: lindechunines A(41) and B(43) were isolated from roots of Lindera chunii Merr., together with 1 new naturally occuring lindenanolide H(31) and 14 known compounds: pseudoneoliderane(32), lindeneol (33), hernandine (34), (+) - heraangerine(35), ocokryptine (36), hernandonine (37), N- methylhernangerine(38), laurolistine (39), 7-oxohernagine(40), 7 -oxohernangerine(42), compounds 2A, 2B, 3 and 4.A simple RP-HPLC method was developed to quantify proanthocyanidin A type trimers (compounds 25 and 26) presented in the stems and roots of L aggregata (Sims) Kosterm..The isolated compounds were tested for their inhibitory activities on HIV-1 integrase (IN), except for some tannin compourds, alkaloid compounds 37, 39, 41 and 42 exhibited significant inhibitory activities with ICy, values of 16. 3, 7.7, 21.1 and 18.2u M, respectively. Structure and activity relationships suggested methylenedioxy group in 7-oxoaporphine alkaloids may play a key role against HIV-1 IN.At last, studies of distribution and mophology of two plants as well as microscopic characteristics of its leaves and roots were carried out.
