喜树热激反应的分子生态学研究

【摘要】 本文以分子生态学思想,特别是生态适应与生态进化的分子机制思想为指导,综合生理生态学、植物资源学与分子生物学研究方法,特别是运用功能蛋白质组学理论与手段,对人工培育的喜树当年生幼苗进行了热激反应的分子生态学研究。 本文研究工作测定了喜树幼苗在热激处理下光合能力的变化,以及MDA、叶绿素等生理指标的动态变化,并测定了不同热激处理条件下喜树体内重要的生物碱——喜树碱和羟基喜树碱的代谢变化。对于不同处理条件,在变性凝胶电泳得到差异条带后,进一步以双向电泳为主要手段,以考马斯亮兰染色法显示蛋白质斑点,共分离得到11个斑点样品,根据肽质量指纹谱检测结果初步鉴定了其中的6个斑点,其中包含7种蛋白质,进一步的生物信息学确认了其中5种蛋白质在喜树热激反应中的生态适应功能。 本文还进一步探讨了喜树碱类物质的热激代谢调控分子机理,在此基础上,结合蛋白质数据,分析了喜树热激反应的生态适应性,并进一步总结了喜树热激反应的分子生态学机理。同时,本文还以喜树对热逆境为模式,探讨了喜树碱代谢调控对喜树抗旱性的可能机制。
【Abstract】 Camptotheca acuminata is a tree unique to China, producing terpenoidindole alkaloid (TIA) 10-Hydroxycamptothecin (HCPT) and camptothecin (CPT), which possess strong anti-cancer bioactivity. The molecular ecology of heat shock to Camptotheca acuminate seedlings with the theory and method of functional proteomics was systematically studied. By the study we attempted to elucidate the molecular mechanism of ecological adaptation and evolving of plant to heat shock in the method of physiological ecology, molecular biology of plants.The research measured the photosynthetic potential changes when the Camptotheca acuminata seedlings were treated by heat shock, at the same time the physiologic index dynamic changes were measured as follows, they were MDA, chlorophyll and so on. We also measured the metabolized changes about the two important alkaloids of Camptotheca acuminata. Under the different treated conditions, differential bands were obtained through the denatured gel electrophoresis, then mainly according to the 2-D Electrophoresis, the dots appeared by the coomassie brilliant blue dying, thus 11 dots were separated. Of these dots, 6 including 7 kinds of proteins were primarily identified in term of the peptide quality fingerprinting map. Then, 5 kinds of the 7 proteins were diagnosed on their ecological acclimation function during the heat shock response by the biological informatics.The study discussed the molecular mechanism on heat shock metabolism regulation of camptothecins. On this base, combined with the protein data, the ecological acclimation of Camptotheca acuminata to heat shock response was analyzed. Then we concluded the molecular ecological mechanism of Camptotheca acuminata to heat shock response. Simultaneously, we used the Camptotheca acuminata heat stress as mode, probed into the possible mechanism on Camptotheca acuminata drought resistance modulated by camptothecinmetabolism.
