脑力苏胶囊改善血管性痴呆大鼠认知障碍作用机制的实验研究

【摘要】 目的:研究脑力苏胶囊改善VD大鼠认知障碍的作用机制。方法:根据Pappas BA方法,采用双侧颈总动脉结扎法制作VD大鼠模型,实验大鼠随机分为假手术组、模型组、喜得镇组、脑力苏胶囊大、中、小剂量组,治疗7天后用水迷宫及跳台实验检测学习、记忆情况;借助光镜、免疫组化方法等对海马组织形态学、血浆降钙素基因相关肽(GGRP)、内皮素(ET-1)含量、脑组织乙酰胆碱酯酶(AchE)含量、海马一氧化氮合酶(NOS)、生长抑素(SS)阳性细胞数进行观测。结果:喜得镇及脑力苏大、中剂量灌胃治疗后大鼠学习记忆明显改善(P<0.01),血浆ET含量明显降低,GGRP含量明显增高(P<0.01);海马神经元密度、AchE含量、SS活性显著升高,NOS活性明显降低(P<0.01)。结论:脑力苏胶囊能显著改善VD大鼠学习记忆能力。其可能作用机制为调节脑缺血时血浆ET-1、CGRP水平, 减轻NOS的神经毒性作用并抑制由此诱发的细胞凋亡;刺激SS分泌,促进胆碱能系统功能。

【Abstract】 Objective: To study the action mechanism of NLS capsule which can decrease ischemia agnosia for vascular dementia rats. Methods: Vascular dementia model rats were established by a 2-vessel occlusion(2-VO) method. These vascular dementia rats were randomly classified into six groups, namely, model group(M),control group(X),NLS capsule high group (Nl) NLS capsule middle group (N2) NLS capsule low dos group(N3) and pseudo-operation group(J).After 7 days treatment, water maze were used to test the learning and memory abilities of these Wistar rats. Observe the pathologocial changes of their cerebral tissues, determine the content of CGRP in serum and AchE in hypomnesia,studied the positive SS and NOS neurons in hypomnesia by immunohistochemical method. Results: From this experiment, the following results could be observed. First, the learning and memory abilities of rats in X group Nl and N2group were better than that of other group rats(P < 0.01).Content of endothelin(ET-1) decreased in serum .otherwise, serum CGRP and AchE of hypomnesia increased( P < 0.01) . Somatostatin (SS) activities ascented by large margin, NOS levels reduced remarkably (P<0.01) .Conclusions: NLS capsule could inhance the learning and memeroy abilities of vascular dementia rats, the mechanism of NLS capsule may be : regulate the level of CGRP and ET-1 in serum, inhibiting expression of NOS which aroused cerebral apotosis, stimulating cerebral tissues to make out SS, raising the function of Ach system.
